Association of interleukin-10 polymorphisms with chronic hepatitis C virus infection in a case-control study and its effect on the response to combined pegylated interferon/ribavirin therapy. Khan AJ, Saraswat VA, Choudhuri G, et al. Epidemiol Infect. 2014 Mar 18:1-10. [Epub ahead of print] http://www.ncbi.nlm.nih.gov/pubmed/24641916 SUMMARY We conducted a case-control study involving 150 genotype 3 chronic hepatitis C virus (HCV) patients and 150 healthy controls to investigate the association of polymorphisms in the interleukin-10 (IL-10) gene with chronic HCV infection and the association of these polymorphic variants with the combination of pegylated interferon (Peg-IFN) and ribavirin therapy response. Our data revealed that the GG genotype of IL-10 -1082A/G exhibited significant association with genotype 3 chronic HCV infection compared to controls. Treatment response data also showed a significant increase in risk for the GG genotype of IL-10 -1082A/G in response-relapse patients or non-responder patients compared to sustained virological response patients. Further, a significant increase in risk was also revealed for the CC genotype of IL-10 -592A/C in response-relapse patients or non-responder patients compared to sustained virological response patients, suggesting a role of the GG genotype of IL-10 -1082A/G and CC genotype of IL-10 -592A/C in the treatment outcome of combined Peg-IFN/ribavirin therapy.

Factors influencing therapeutic response to peginterferron plus ribavirin in the different genotypes of chronic hepatitis C

Abstract Background: Different factors like age, sex, route of infection, initial viral load, levels of liver function tests and genotypes may affect response to treatment in patients with chronic hepatitis C. The purpose of this study was to determine the role of these factors in the treatment of these patients. Methods: From 2004 to 2012, different genotypes of chronic HCV (Hepatitis C ...

Polymorphism of IL-28B Gene (rs12979860) in HCV Genotype 1 Patients Treated by Pegylated Interferon and Ribavirin

Background: Nowadays, the immune response to hepatitis C (HCV) treatment has become a crucial issue mostly due to the interleukin 28B (IL-28B) polymorphism effects in chronic HCV patients. The aim of this study was to detect the polymorphism of IL-28B gene (rs12979860) in HCV genotype 1 patients treated with pegylated Interferon and Ribavirin. Methods...

Pharmacogenetics of hepatitis C: transition from interferon-based therapies to direct-acting antiviral agents

Hepatitis C virus (HCV) has emerged as a major viral pandemic over the past two decades, infecting 170 million individuals, which equates to approximately 3% of the world's population. The prevalence of HCV varies according to geographic region, being highest in developing countries such as Egypt. HCV has a high tendency to induce chronic progressive liver damage in the form of hepatic fibrosis...

بر IL 28B ژن rs اثر پلی مورفیسم تک نوکلئوتیدی 12979860 پاسخ به درمان با پگ- اینترفرون/ریباورین در بیماران C ایرانی مبتلا به هپاتیت

Background and purpose: The standard of care treatment for infected patients with HCV is based on a combination of pegylated interferon alpha and ribavirin. Recently, the rs12979860 SNP polymorphism, located upstream of the interleukin 28B gene, was shown to be strongly associated with response to anti-HCV therapy. This study investigated the distribution of the (C/T) polymorphism with sust...
